SCTB: Vector Production

Overview of Services

The vector core assists investigators with design, construction and/or production of viral vectors. We have a turnaround time of approximately two to three weeks between receiving the transfer plasmid and final product release.

Our services include:

Retroviral Vector Production and related Services (Gammaretro, Lenti, Foamy)
· 2nd generation high-quality, high titer laboratory grade production (BSL-2)
· 3rd generation high-titer lentiviral laboratory grade production: approved by many academic institutes (e.g. University of Washington) for in vivo applications and BSL-1 application without prior test for replication competent virus (RCL assay)
· "From Scratch" service: transfer plasmid design and assembly following your needs and specifications
· RCL assay: testing for replication competent lentivirus (RCL) in lentiviral vector products of required by IRB/EH&S (core-produced vector only)
· quantification of functional units by flow cytometry (fluorescent transgene/antibody stain)
· quantification of functional vector units by Taqman qPCR (lenti IU/ml)

AAV Production and related Services
· High-quality high titer production (laboratory grade)
· Fast, cost effective vector production for in vitro applications
· Large scale production of concentrated, purified vector for in vivo applications (AAV2, AAV6 only)
· Quantification of functional units by flow cytometry (fluorescent transgene/antibody stain)
· Quantification of functional vector units by Taqman qPCR (GC/ml)

Ready-to-use vector aliquots for immediate pick-up
· Aliquots of common AAV & retroviral vectors titered and tested for functionality
